WP01 - COORDINATION OF SCIENTIFIC AND TECHNICAL STRATEGY
The objective of work package WP01 is to ensure implementation of a consensus-based strategy and to monitor and adjust project execution.
WP02 - BIOBANKS AND SOURCING
The aim of work package WP02 is to constitute a first collection of 20 venoms and venom glands that will be used to set up proteomics and transcriptomics sequencing protocols for WP03 and WP04. Emphasis will be placed on sample biodiversity, with the sourcing of arachnid, insect, snake, cone and cnidarian specimens through missions in Europe, Metropolitan and overseas French territories.
WP03 - PROTEOMICS RTD: HIGH-THROUGHPUT DE NOVO PEPTIDE SEQUENCING
The goal of work pacakage WP03 is to generate a database of ca. 4,000 peptide sequences based on the investigation of the first collection of 20 venoms from WP02 using cutting edge mass spectrometry instruments and advanced gas-phase activation methods. Novel peptide sequencing techniques based on chemical derivatization will be implemented and specific sequencing interpretation software will be developed for the automated analysis of data.
WP04 - TRANSCRIPTOMICS RTD: HIGH-THROUGHPUT cDNA SEQUENCING
Work Package WP04 is dedicated to the setup of transcriptomics sequencing using the venom glands collection from WP02. Various RNA extraction protocols will be compared and sequencing will be performed on different next-generation platforms. WP04 will also involve construction of the VENOMICServer, the database that will centralize all sequences generated by proteomics and transcriptomics sequencing.
WP05 - PRODUCTION RTD: HIGH-THROUGHPUT PARALLEL PEPTIDE PRODUCTION
The setup of high-throughput peptide production from database sequences, which is the objective of work package WP05, will involve optimization of production strategy and protocols through the synthesis of a set of 200 known toxins and 200 novel toxins selected from WP04. Peptides will be synthesized using both recombinant production in Escherichia Coli and Solid-Phase Peptide Synthesis techniques. Proper folding of toxins will be ensured through the use of a proprietary platform already developed. Quality controls of peptides produced will be performed by mass spectrometry, Nuclear Magnetic Resonance and pharmacological assay.
WP06 - VENOMICSERVER DATABASE
Work package WP06 aims at developing the project database that will centralize, in addition to sequences generated by proteomics and transcriptomics, all information and datasets produced by the VENOMICS project. VENOMICServer will come under the form of a controlled-access web database allowing data query, visualization and analysis. It will be connected to public biological databases for DNA/Protein sequences (NCBI, Uniprot, Prosite, Pfam), protein structure (Protein Data Bank, PDB), literature references (Pubmed) and biological categories (Gene Ontology).
WP07 - PEPTIDE BANK
The objective of work package WP07 is to generate a 10,000 peptide bank by high-throughput automated peptide production using the strategy optimized in WP05. The workpackage will include the identification of molecular targets representing unmet therapeutic needs and potential markets, and the screening of the peptide bank against these targets to isolate and validate drug leads.
WP08 - COMMUNICATION AND DISSEMINATION OF RESULTS, IP MANAGEMENT, BUSINESS DEVELOPMENT
Work package WP08 will focus on the management of knowledge generated by the project, including external communication, protection and valorization at the business level. The main objectives are to give wide visibility to the project toward the outside world, and setting the foundations for future commercial exploitation and opportunities.
WP09 - PROJECT MANAGEMENT
Work package WP09 will be dedicated to the implementation of a Project Management Office (PMO) in charge of monitoring and adjustments of project schedule, budget, or administrative tasks.
